Tips for keeping your Applikon® Bioreactors in good working order:
To clean the sensors:
Drain the reactor after finishing the cultivation
Remove the pH and DO sensors, clean both sensors thoroughly. Make sure that no remainders of the broth are left behind on the sensor surface. Store the pH sensor in water or, to expand its lifetime, in a KCl solution (c = 3M) (The DO sensor may be stored dry.).
To clean auxillaries and tubing:
Disassemble the bioreactor system and remove all tubing and auxiliaries that are inserted in the reactor (side wall and head plate). These assemblies must be cleaned “off-place” with the same cleaning solutions as were used for thereactor.
Rinse thoroughly with tap water (3 times).
Rinse with de-ionized water (or Water For Injection / High Purity Water).
Check the quality of O-rings and gaskets during reassembling. Replace them if necessary.

Applikon® Bioreactors Troubleshooting Tips:
Verify Sensor Calibration and Signal Integrity:
Before interpreting unexpected process values, confirm that all sensors—particularly pH, dissolved oxygen, and temperature probes—are correctly calibrated and providing stable signals. Drift, electrical noise, or poor electrode maintenance can often exhibit process deviations.
Inspect Control Loops for Proper Tuning:
If the system exhibits oscillations or fails to reach setpoints efficiently, review PID tuning parameters for each control loop. Overly aggressive or sluggish control behavior often points to improper tuning rather than mechanical failure.
Confirm Gas Flow and Mixing Uniformity:
Irregular DO or pH readings may result from inadequate gas sparging or mixing inefficiency. Check for clogged spargers, leaks in gas lines, or improper impeller speed settings to ensure homogeneous mixing as well as mass transfer.
Assess Software Configuration and Communication Links:
Unexpected control responses or data loss may stem from software misconfigurations or communication errors between the controller and peripheral devices. Verify firmware versions, communication protocols, and ensure that all device addresses are correctly assigned.
Evaluate Sterility and Mechanical Integrity Before Each Run:
Inconsistent culture performance often originates from undetected leaks or contamination. Inspect vessel seals, sampling ports, and tubing connections for wear or residue.
