Tips for keeping your AKTA™ Prime Plus in good working order:

  • Keep the UV flow cell clean. Do not allow solid solutes to aggregate in the flow cell, and ensure no solid particles enter the flow cell.

  • Flush the liquid flow path after use with purified water and 20%ETOH.

  • Calibrate the pH electrode before each use. The pH electrode should be stored in a 1:1 mixture of pH 4 buffer and 2 M KNO3 when not in use

  • Check the inlet filters weekly. Do the same for the on-line filter if your system has one.

  • Check that the flow restrictor generates the appropriate back-pressure of 0.4 ± 0.05 MPa. This should be done monthly.

  • Inspect all valves and check for leaks regularly.

  • Routinely clean all flow paths to prevent growth or contamination. If you have growth, that means you haven’t been cleaning your system properly! Proper cleaning should be a prevention, not a remedy.

  • Maintain a stable ambient temperature and low ambient humidity. Low temperatures and high humidity lead to corrosion!

AKTA™ Prime Plus Troubleshooting Tips:

  • Most problems are caused by the system being dirty or having air in it. When in doubt, clean the system and purge the lines. The UV cell should be cleaned with Decon™ 90, Deconex™ 11 or similar cleaners, while the conductivity cell should be cleaned with 1 M NaOH or 20% ethanol.

  • Ghost peaks are usually contamination or residue from previous runs. Clean the system and column, ensuring air is removed. It could also be a problem with the mixer. Ghost peaks in the gradient profile, however, are usually due to air in the flow cell. Check for loose connections and use the flow restrictor.

  • A poor UV signal could also be contamination, but it could also be impure buffer or air. Purge the pump, run a system wash, and test the signal with ultrapure water to see if it gives a clean signal.

  • Low sensitivity is often a sign of an aging UV lamp which needs replacement. However, it could also be that the UV lamp and filter are not both set to the wavelength being used.

  • Baseline drift may be caused by leaks, improper mixer function, or a non-equilibrated column.

  • Wavy gradients may be due to pump problems, although it can also be due to the mixing chamber. Contamination, insufficient chamber volume, and mixing motor failure can all be causes.
